BINDING ASSAY USING A RADIOLABELLED (125I) LIGAND

This assay is used for measuring the binding of proteins to whole cells or cell membranes and assessing their binding affinity and specificity.

PRINCIPLE

The assay allows characterisation of the interaction of a protein of interest with cell membrane-associated binding partners. Whole cells or isolated membrane fractions can be used for this purpose. The protein to be tested is labelled with radioactive iodine (125I). Bound radioactivity is measured by a gamma-counter. The data obtained are used to determine binding parameters such as Kd (dissociation constant) and Bmax (number of binding sites per cell).

RESULTS

Results are calculated using specialised data analysis software (GraphPad Prism).  Nonlinear regression analysis is performed on the obtained binding curve to determine ligand affinity (Kd) and other binding parameters.
